Offer Description
Effects of artificial KCC2 clustering on intracellular chloride regulation in hippocampal interneurons and pyramidal cells, and on epileptic activity
We will develop super-resolution imaging and single-molecule tracking of KCC2 in interneurons and pyramidal cells in mouse hippocampal slices or human epileptic tissue.
After characterizing and comparing the membrane distribution and mobility of KCC2 in interneurons and pyramidal cells, we will determine the impact of antibody-induced artificial KCC2 clustering.
We will then investigate the functional impact of this strategy using electrophysiology (patch-clamp recordings of EGABA and multielectrode array (MEA) recordings of network activity) in human epileptic tissue slices.
The successful candidate will join the La Synapse Dynamique team, led by Sabine Lévi and based in the Brain Plasticity Unit (UMR 8239) at ESPCI Paris. Our research focuses on the cellular and molecular mechanisms underlying the stabilization of nascent synapses during development and the plasticity of mature synapses, with a particular emphasis on hippocampal GABAergic synapses. We use single-molecule imaging techniques, including single-molecule tracking in live neurons and super-resolution STORM/PALM microscopy.
